چکیده

It was investigated to clarify the relationship between the composition of the lipid fractions obtained from the ascitic fluid of Ehrlich ascites tumor bearing mice and its uncoupling activity after whole body irradiation (1,000 r). 1. Oxidative phosphorylation of Ehrllch ascites tumor cells was loosely uncoupled with the addition of ascitic lipid fraction extracted from tumor bearing mice. 2. The uncoupling activity of the lipid fraction on the oxidative phosphorylation of the tumor cells increased after whole body irradiation. 3. Ascitic lipid fraction, especially acetone soluble fraction accelerated mitochondrial swelling, and the swelling action was increased remarkably by the whole body irradiation. 4. No significant changes were observed in the proportion of acetone soluble fraction to acetone insoluble fraction in the ascitic lipid after X-irradiation, and the proportion of the both fractions was approximately 9 : 1, respectively. 5, Main compositions of total and nonesterified fatty acids in the ascitic fluid obtained from the control and X-irradiated groups were palmitic, stearic, oleic, linoleic and palmitoleic acids, and the proportions of unsaturated acids, especially oleic and linoleic acids in both fatty acid fractions were greater in the X-irradiated group. 6. Remarkable increment of unsaturated fatty acid especially linoleic acid, was also observed in the total fatty acids of the tumor cells separated from the X-irradiated group. 7. It can be concluded that an uncoupling agent extracted from ascitic fluid of the X-irradiated group was a mixture of long-chain fatty acids, especially oleic and linoleic acids. 8. It was also discussed that uncoupling oxidative phosphorylation in liver mitochondria after whole body irradiation may be caused by a similar mechanism to that in the turnor cells. ∗PMID: 14201061 [PubMed indexed for MEDLINE] Copyright c ©OKAYAMA UNIVERSITY MEDICAL SCHOOL Acta Med. Micro-pixe analysis in invasive ductal carcinoma tissues after treatment of astaxanthin

Background: Trace elements play an important role in a number of biological processes. Astaxanthin (ASX), a carotoid pigment found in certain marine plant and animals, has shown anti cancer and anti free radical properties. This work intended to understand the effect of Astaxanthin in breast cancer (invasive ductal carcinoma, IDC) by using micro-pixe method. For this aim the concentratio...

Identification of serpin (alpha-1-antitrypsin) as serum growth inhibitory factor in murine ehrlich carcinoma by proteomics.

It is well established that serum factors play a role in relapse of tumor diseases after removal of the primary tumor. The molecular nature of these factors and their mechanism of action remain unknown. We focused on host-related mechanisms to identify tumor-specific serum factors of mice bearing Ehrlich carcinoma, which have the potential to confer resistance towards tumor development. An expe...

Use of raw Euphorbia tirucalli extract for inhibition of ascitic Ehrlich tumor.

OBJECTIVE to evaluate the effect of the Euphorbia tirucalli hydroalcoholic extract (ETHE) on the development of Ehrlich Tumor, in its ascitic form. METHODS we intraperitoneally inoculated 15 Swiss mice with 10.44 x 107 cells of Ehrlich Tumor and divided them in two groups one day after: ETHE Group (eight mice), treated with a dosage of 125 mg/kg/day of EHTE for five days; and Control Group (s...

Gold nanoparticle-induced sonosensitization enhances the antitumor activity of ultrasound in colon tumor-bearing mice

Introduction: Light-driven cancer therapy strategies (e.g. photodynamic therapy and photothermal therapy) have undergone remarkable progress in recent years, but they still suffer from a serious drawback of limited penetration depth of light in tissue. As a non-invasive and non- ionizing radiation, ultrasound can be focused remotely, transferring acoustic energy deep in the bo...

Antitumor activity of silver nanoparticles in Dalton’s lymphoma ascites tumor model

Nanomedicine concerns the use of precision-engineered nanomaterials to develop novel therapeutic and diagnostic modalities for human use. The present study demonstrates the efficacy of biologically synthesized silver nanoparticles (AgNPs) as an antitumor agent using Dalton's lymphoma ascites (DLA) cell lines in vitro and in vivo. The AgNPs showed dose- dependent cytotoxicity against DLA cells t...
